Выявлено, что желобковое соединение H33258, специфичное к дц-структуре нуклеиновых кислот, может связываться и с оц-РНК. Обнаружено, что H33258 лучше взаимодействует с образовавшимся в результате гибридизации полирибонуклеотидом poly(rA)-poly(rU) по сравнению с оц-poly(rA) и оц-poly(rU). На основании спектров флуоресценции построены кривые связывания H33258 с оц- и дц-полирибонуклеотидами и определены константы связывания K и число оснований n, приходящихся на одно место связывания. Получено, что константа связывания H33258  с оц-poly(rA) в 4—5 раз больше, чем с оц-poly(rU). Выявлено, что сродство H33258 к poly(rA)poly(rU) и оц-poly(rA) зависит от ионной силы раствора, в то время как в случае оц-poly(rU) такая зависимость не обнаружена. </p></abstract><trans-abstract xml:lang="en"><p>Comparative study on the interaction of Hoechst 33258 (H33258) with single-stranded (ss-) polyribonucleotides poly(rA) and poly(rU) and double-stranded (ds-) poly(rA)-poly(rU) has been carried out with the method of fluorescence spectroscopy. Groove-binding compound H33258, being specific ligand for NA ds-structure, was revealed to bind to ss-RNA. It was revealed that H33258 shows higher preference to poly(rA)-poly(rU), formed due to the hybridization, than to ss-poly(rA) and poly(rU). Based on the fluorescence spectra the binding curves of H33258 to ss- and ds- polyribonucleotides were constructed and the values of binding constant K and number of bases per binding site n were determined. The obtained data elucidate that the binding constant of H33258 to ss-poly(rA) is 4-5 times higher, as compared to ss-poly(rU) one.
